Sample preparation Weigh 2.5 g of the powdered sample and place it in a 50-mL centrifuge tube, then add 10 mL of methanol (60%). Sonicate (350 W) the mixture for 15 min. Centrifuge at about 4000 × g for 5 min. Filter through a 0.45-µm nylon filter
Mobile phase Ethyl acetate, ethanol, formic acid, water 18:3:1.5:1 (v/v/v/v)
Derivatization No derivatization
Sample preparation Weigh 1.0 g of the powdered sample and place it in a 50-mL conical flask, then add 10 mL of methanol. Sonicate (200 W) the mixture for 15 min. Filter the mixture
Mobile phase Ethyl acetate, water, formic acid, glacial acetic acid 8:2:1:1 (v/v/v/v)
Derivatization Spray with 1% vanillin sulfuric acid reagent in ethanol, and heat at 105°C until the sppots appear
