Sample preparation To 0.5 g of the powdered herbal drug add 5 mL of a mixture of equal volumes of ethanol (96%), water and heat to boiling. Centrifuge and use the supernatant liquid
Mobile phase Acetic acid, water, ethyl acetate, propanol 1:30:40:40 (v/v/v/v)
Derivatization Treat with a 20% v/v solution of nitric acid and heat at 120 °C for 10 min. Allow to cool and treat with a 50 g/L solution of potassium hydroxide in ethanol (50% v/v) until the zones appear
