Sample preparation Dilute 10 µL of the essential oil to be examined to 1 mL with toluene
Mobile phase Toluene, ethyl acetate 95:5 (v/v)
Derivatization Treat with anisaldehyde solution, heat at 100 °C for 3 min; examine immediatly in daylight
System Suitability Test (SST) Under UV 254, prior to derivatization
  • Isoeugenol: quenching zone at RF ~ 0.36
  • Isoeugenyl acetate: quenching zone at RF ~ 0.40
Specific marker(s) Under UV 366 nm, after derivatization
  • 6-gingerol: brownish-purple zone RF ~ 0.02
  • geraniol: purple zone RF ~ 0.13

Sample preparation To 1.0 g of the powdered herbal drug add 10 mL of methanol. Sonicate for 10 min and centrifuge
Mobile phase Toluene, ethyl acetate 3:1 (v/v)
Derivatization Derivatize with anisaldehyde reagent, and heat the plate at 100°C for 3 min.

  • 6-gingerol: orange zone RF ~ 0.24
  • 8-gingerol: orange zone RF ~ 0.27
